The emergence of multi drug resistant pathogens such as Methicillin Staphylococcus aureus (MRSA) has become a threat to public health. Thus, the development new antibiotics global concern. Many scientists are attracted towards discovery antimicrobial agents from microbial sources. aim this study is isolate potential Bacillus species soil for production substance. During study, 20 samples were collected various areas Kathmandu valley. A total 70 isolates identified by morphological and biochemical characteristics. Altogether 39 able produce antibacterial extract when it was grown in Trypticase soya broth (TSB) at 37°C 48 h. activity tested against test organisms like Escherichia coli, Klebsiella pneumoniae, Salmonella, Pseudomonas, Candida Agar well diffusion method. All extracts showed zone inhibition E. coli except 6 (E39, E43, E36, E25, E45 E46). highest shown B68 with 11 mm. Only one (E62) inhibit growth Pseudomonas. (E35) S. (6mm). protein nature determined Biuret Ninhydrin tests. relatively stable heat treatment unstable surfactants 70% methanol, 2-propanol ethylacetate. revealed that good source producer bacteria.
